Lab 7 Evaluation of muscle relaxation action of Diazepam using Rotarod

Aim of practical: Purpose of this experiment is to evaluate the muscle relaxation action of Diazepam using Rotarod.

Theory: Diazepam, a benzodiazepine group of drug, possesses multipose activities like CNS depression, anti convulsion, sedative and hypnotic action, anti epileptic action, anxiolytic action, muscle relaxation activity etc. It acts by increasing the neuronal membrane permeability to chloride ions by binding to stereo-specific benzodiazepine receptors on the postsynaptic gamma-amino butyric acid (GABA) neuron within the CNS and thus enhancing the GABA inhibitory effects resulting in hyper polarization and stabilization [http://www.mims.com]. It produces muscle relaxation action probably by inhibiting the polysynaptic afferent pathway.

Clinical uses: Major motor seizures, Status Epileptics, Acute anxiety Skeletal muscle relaxant, Muscle spasm, Alcohol withdrawal symptoms, Vertigo, Spastic disorders, Insomnia, Pre anesthetic medication, Tetanus. The onset of action is one to five minutes for IV administration and 15-30 minutes for IM administration. The duration of diazepam's peak pharmacological effects is 15 minutes to one hour for both routes of administration. Peak plasma concentration is achieved after 30-90 min by oral route. Elimination half-life is biphasic: Rapid (initial): 1-2 days; terminal: 2-5 days.
